Flow Cytometry based 'Acute Myeloid Leukemia Maturity Score' is a Novel Marker for Predicting Relapse in Acute Myeloid Leukemia: A Prospective Observational Study

Objectives: A novel flow cytometric ‘AML Maturity Score (AMS)’ classifying patients into AML-immature (AML-im) and AML-mature (AML-ma) based on CD34, CD117 Tdt expression blasts correlated with induction remission, relapse free survival overall in previous studies. We aimed to study the correlation between AMS ELN risk group outcomes- remission (IR), time (TTR) relapse.
 Material Methods: This was a combined prospective retrospective of 104 AML over 30 months. Flow cytometry quantitative TdT used for calculating additional tube CD34+CD38-CD123+ cells put at diagnosis 47 prospectively studied. The received 1-2 cycles standard (3+7) comprising Daunorubicin (60mg/m2/day 3 days) Cytarabine (continuous infusion 100mg/m2/day 7 days). Data collected clinical laboratory parameters, cytogenetics molecular profile these who were followed up IR, TTR Results: Of patients, AML-im 86.6% (90/104) AML-ma 13.4% (14/104). Patients classified as – favorable (28.8%), intermediate (50%) adverse (21.1%) basis 2017. 74 attained IR 65 mean period 18 months, 27 relapsed. significantly more (p=0.003). On AML-ma, (p=0.000) relapses (p=0.043) higher group.
 Conclusion: is potential marker predicting patients.
